Kaja Helvik Skjærven has a special interest area related to early embryonic development in fish and how development is affected by environmental conditions such as nutrition, temperature, or toxicants. The core focus is how the different environmental conditions change the potential for gene expression through nutritional based epigenetic changes. The main theme of the work is how nutrients, temperature, or toxicants regulate specifically DNA methylation (epigenetics) intergenerationally (from parents to offspring) and associates the changes in gene expression potential (epigenetics) with more traditional biological procedures like nutrition, physiology and histological analyses.

 

A few years ago, we wanted to test whether parental vitamin status could affect the next generation. We made a feed with lower vitamin level and gave it to the parents. It lowered the fecundity, but the eggs that were spawned were without deformities. However, RNA sequencing revealed that the embryo stage was affected by parental vitamin status (Skjærven et al., 2016, https://www.nature.com/articles/srep34535). In the next generation, both groups were fed the control feed with sufficient B-vitamin level which resulted in equal growth of the F1 generation. We were quite surprised when we open to dissect the liver of the children, they had pale pink liver color, and the histology showed significantly more fat in the liver, down-regulated genes in fat metabolism and changed over 2800 differentially methylated sites. This is nutritional programming and DNA methylation was one of the mechanisms that controlled the development of the phenotype (Skjærven et al., 2018, https://www.nature.com/articles/s41598-018-21211-5). Recently, we have also shown that changing only one nutrient in the parental feed influenced the which genes where expressed and DNA methylation in the offspring livers (Adam et al., 2019).
